Spyre Therapeutics, Inc. (NASDAQ:SYRE) stock is trading lower on Wednesday after the company shared topline findings from the rheumatoid arthritis (RA) segment of its Phase 2 SKYWAY trial for SPY072.
While the drug demonstrated statistical significance in improving symptoms and exhibited a solid safety profile, the results fell short of the company’s internal bar to prioritize advancement of SPY072 in RA.
Efficacy And Target Engagement
Trial data released on Tuesday indicated that both SPY072 doses successfully hit target drug concentrations. The therapy provided complete, durable suppression of free TL1A through 12 weeks, signaling complete target engagement.
The low dose achieved a statistically significant advantage over a placebo on the primary endpoint, measuring the change from baseline in DAS28-CRP at Week 12.
Furthermore, the high dose generated nominally significant improvements for the secondary ACR20 endpoint, while the low dose achieved similar success on the exploratory ACR50 endpoint.
Researchers noted comparable outcomes between patients new to advanced therapies and those with prior exposure.
Safety Profile And Future Strategy
Participants tolerated SPY072 well. The safety profile aligned with expectations for the TL1A class. The treatment arm recorded adverse events at a rate of 27%, compared to 36% for the placebo group.
The company plans to deliver topline data for SPY072 in psoriatic arthritis and axial spondyloarthritis by the fourth quarter of 2026 and for SPY072 in combination with IL17-A/F in hidradenitis suppurativa in late 2027 or early 2028.
