On Wednesday, the U.S. Food and Drug Administration (FDA) approved Takeda Pharmaceutical Co. Ltd.‘s (NYSE:TAK) Orzeyful (oveporexton), an oral orexin receptor 2 (OX2R) agonist, for narcolepsy type 1 (NT1, narcolepsy with cataplexy) in adults.
This oral orexin receptor 2 agonist marks the first treatment of its kind designed to address the condition comprehensively rather than just managing specific symptoms.
Comprehensive Approach To Narcolepsy Type 1
Narcolepsy type 1 is a rare neurological disorder caused by an orexin shortage. It currently impacts roughly 120,000 individuals in the U.S.
Patients often suffer from extreme daytime drowsiness, sudden muscle weakness known as cataplexy, cognitive challenges, and fragmented nighttime rest, which can severely disrupt daily activities like work and social engagements.
The regulatory clearance stems from robust data gathered during the global Phase 3 FirstLight and RadiantLight trials.
These studies demonstrated that oveporexton provided statistically meaningful enhancements across a broad spectrum of symptoms.
The FDA, in a press release, noted that rather than masking symptoms with stimulants or sedatives, Orzeyful directly activates the same brain receptor that the body’s own orexin would normally stimulate, restoring the missing signal.
Future Availability
Currently, the Drug Enforcement Administration is assessing the drug for controlled substance classification, a process anticipated to conclude within 90 days.
Following this determination, the medication will roll out to healthcare providers and patients via a specialty pharmacy.
